The Linnet study, which evaluated the bispecific antibody in patients with advanced clear cell gynecologic cancer, produced a 25% confirmed objective response rate. Despite these results, the trial data revealed that 49% of participants suffered Grade 3 or higher treatment-related adverse events. Clinical complications reached a critical threshold with the death of one participant in the platinum-resistant ovarian cancer cohort.
Common side effects included anemia, neutropenia, and elevated aspartate aminotransferase levels. Faced with these safety concerns and a 24% dropout rate, MacroGenics confirmed plans to test lower dosages in a new cohort. The company intends to share updated findings in the first half of 2027. While a 44% disease-control rate was observed, with all responding patients maintaining their status as of the September 1 cutoff, the market reaction reflects investor anxiety regarding the drug's toxicity profile.
